paroF->MCherry-MutH(E56A)
This biobrick contains mutH(E56A) and mCherry under the control of the tyrosine-sensitive promoter paroF. Unless the E. coli cell produces very high levels of tyrosine, the mutagenizing protein mutH(E56A) and mCherry will be produced. The mCherry is used as a reporter to indicate when this maximum tyrosine production is reached (florescence will decrease). mutH should cause genomic mutations that should result in the cell eventually producing high levels of tyrosine.
